AVALIAÇÃO HISTOLÓGICA PARA APÊNDICES LOCOMOTORES DE NEOHELICE GRANULATA AVALIAÇÃO DA EFICÁCIA DE FIXADORES, DESCALCIFICADORES E CORANTES PARA ESTUDO DE APENDICES LOCOMOTORES DE NEOHELICE GRANULATA (DANA, 1851)

Foram coletados dezesseis espécimes de Neohelice granulata no estuário da Laguna dos Patos sendo que, de cada um
destes foram coletados cinco apêndices locomotores. Dois destes apêndices foram fixados em Formol a 10%, outros dois em
Zenker e um em Davidson, correspondendo a 32, 32, 16 repetições respectivamente. Após os apêndices terem sido fixados em
Formol e Zenker estes foram colocados nos descalcificadores Ácido Nítrico 5% e Solução de Citrato de Sódio/Ácido Fórmico.
Já os apêndices fixados em Davidson permaneceram nesta solução, pois se trata de um fixador/descalcificador. Após o
processamento do material as lâminas foram coradas com a técnica de coloração com Hematoxilina e Eosina (HE), e pela
técnica Tricrômico de Gomori e Tricrômico de Mallory. Através das análises determinou-se que não há diferença estatística
entre os tratamentos, porém sugere-se o fixador e descalcificador Davidson tendo em vista sua facilidade de manuseio e os
baixos custos. Com relação aos corantes observou-se que não houve diferença (P>0,05) entre as técnicas HE e o Tricrômico
de Mallory, no entanto, estes foram melhores (P<0,05) que o Tricrômico de Gomori. Sendo o HE o mais indicado devido ao
preço e facilidade da técnica para o estudo histológico com apêndice locomotor de Neohelice granulata.

There were collected sixteen specimens of Neohelice granulata in the estuary of the Laguna dos Patos (Dos Patos Lagoon).
From each of them, five pereiopods were collected. Two of these pereiopods were fixed in 10% Formol, other two of them in
Zenker and one in Davidson, in a total of 32,32,16 repetitions respectively. After they had been fixated in Formol and Zenker
they were immersed in the decalcifiers 5 % Nitric Acid and in a solution of Sodium Citrate/Formic acid. On the other hand, the
appendages fixated in Davidson remained in this solution because it is a fixative/decalcified. After the material processing the
glass slides were stained with the staining techniques Hematoxylin and Eosin (HE), Gomori Trichrome (TG) and Mallory
Trichrome (TM). Throughout the analysis, there was determined no statistics difference between the treatments, yet it is
suggested the usage of the Davidson fixative/decalcifier taking into account its handling easiness and low cost. Regarding the
colorants, it was observed no difference (P>0.05) between the HE and Mallory Trichrome techniques, however they were found
better than the Gomori Trichrome. The HE is the most indicated due to its price and technique easiness for the histological
study with the Neohelice granulata pereiopod.
